I am fairly new here and I need some advice. I want to start an online printing company and the site should be working and looking like this flyer printing company. Can anyone suggest which kind of e-com component and template I should use in order to get the calculation bit like the one from the site?

Well virtuemart can do the part with options selection and price calculation, but I presume your customers need to upload graphic, you would need to think of an way to do that.

I have seen plugins for upload file on the checkout stage, but my biggest concern is to have the calculation bit. I am not sure if that is possible with virtuemart??

You can have unlimited attributes , whit price add value for attribute value

You can customize your VM flypage and put it as home page and that is it. You would have single product with attributes.
